Resumen: Dravet Syndrome Conference 2023

El pasado jueves 23 de marzo tuvo lugar en Madrid la Dravet Syndrome Conference 2023, organizada por la Fundación Síndrome de Dravet. En un marco incomparable, como es la sede del Colegio Oficial de Enfermería de Madrid, nos reunimos numerosos expertos internacionales, incluyendo profesionales sanitarios, investigadores y profesionales de la industria farmacéutica, con el objetivo común de mejorar la calidad de vida de los pacientes con síndrome de Dravet. Ha sido una oportunidad única para ponerse al día con el manejo de la enfermedad, conocer los últimos avances y tratamientos de esta enfermedad.

Tras la inauguración de las jornadas, la conferencia comenzó el testimonio de Lauraine Mckenna, mamá del pequeño Patrick, que nos contó su experiencia como madre de un niño con síndrome de Dravet, los retos a los que se enfrenta día a día y cómo afecta a la salud mental y física de cada padre y madre, así como sus expectativas y esperanzas. A continuación, la Dra. Charlotte Dravet, que a sus 86 años se desplazó para participar en nuestras jornadas, nos explicó desde su perspectiva cómo ha evolucionado y progresado la enfermedad de un siglo a otro.

Después, disfrutamos de varias sesiones en las que el tema principal fue las comorbilidades que coexisten con el síndrome de Dravet. El Dr. Andreas Brunklaus, del Royal Hospital for Children de Glasgow, nos habló sobre la importancia del diagnóstico e intervención precoz para minimizar el impacto a largo plazo de las comorbilidades, para lo que se ha desarrollado un modelo predictivo que combina la edad de inicio de las crisis epilépticas con la mutación genética en el gen SCN1A y que permite una predicción temprana de si el paciente desarrollará el síndrome de Dravet o una epilepsia genética con crisis febriles, lo cual ayudaría tanto al pronóstico como a la aplicación precoz de terapias. Lieven Lagae, de KU Leuven, se centró en la evolución de los trastornos cognitivos en el síndrome de Dravet y recalcó que suelen aumentar con la edad con muchos factores a tener en cuenta. Comentó también que la administración de fármacos contraindicados tiene un efecto muy negativo en la cognición de estos pacientes, por lo que es clave el diagnóstico precoz. Desde el Hospital Sant Pau Barcelona, la Dra. Susana Boronat nos contó acerca de las comorbilidades neuropsiquiátricas y la importancia de los familiares y cuidadores para entenderlos. El Dr. Franck Kalume de la Universidad de Washington explicó también en su propia charla que la disfunción en los canales iónicos SCN1A es en sí mismo un factor de riesgo para SUDEP, ya que las interneuronas GABAérgicas de varios núcleos del tronco encefálico se verían afectadas por la pérdida de función de estos canales iónicos, lo que conduciría a los cardiacos y respiratorios que podrían causar SUDEP. Para finalizar este ámbito, el Dr. Ángel Aledo-Serrano, del Hospital Vithas Madrid La Milagrosa, culminó con la definición del éxito terapéutico mejorando el bienestar de los adultos.

Tras un breve descanso, siguió la conferencia centrada en el síndrome de Dravet en adultos. La Dra. Estíbaliz Barredo, del Hospital Universitario Gregorio Marañón, habló del camino que recorre el paciente con síndrome de Dravet en España, a través de un proyecto que se ha realizado en colaboración con la Fundación Síndrome de Dravet. Desde el Hospital La Fe, el Dr. Vicente Villanueva nos explicó que los pacientes con síndrome de Dravet suelen presentar en la adolescencia o adultez temprana la marcha agachada, síntomas parkinsonianos que se agravan con la edad, así como alteraciones del habla y lenguaje y problemas para iniciar y mantener el sueño. Remarcó también que las crisis epilépticas mioclónicas, focales y de ausencia se asocian a un peor desarrollo cognitivo. La Dra. Danielle Andrade, desde la Universidad de Toronto, nos contó la transición del paciente teniendo en cuenta las comorbilidades y la necesidad de realizar más estudios en adultos, ya que por cada 7 estudios pediátricos solo se realiza uno en adultos. Para cerrar la sesión, la Dra. Sarah Weckhuysen, desde la University of Antwerp, nos explicó en su charla que hay muchos adultos sin diagnosticar debido a que no tuvieron a pruebas genéticas cuando eran más jóvenes pero que también se debe considerar realizar nuevos tests genéticos en adultos, ya que hay mutaciones y genes que en su día no se tenían en cuenta. Terminó remarcando la necesidad de concienciar a los neurólogos de adultos sobre las epilepsias genéticas y las pruebas diagnósticas genéticas.

Tras un interesante descanso en el que pudimos intercambiar opiniones y pensar en futuras acciones, nuestra directora científica, la Dra. Vittoria Spinosa inauguró la sesión de la tarde con una actualización de los tratamientos en camino para el síndrome de Dravet, remarcando la necesidad de conocer mejor las terapias avanzadas por los padres y cuidadores para tomar decisiones informadas en el futuro. La Dra. Verónica Rivero nos habló sobre el kit de investigación del síndrome de Dravet de acceso abierto que se está desarrollando desde Fundación y la Dra. Lorianne Masuoka, desde Epygenix Therapeutics, nos explicó cómo el nuevo método de alto rendimiento para el cribado de fármacos a través de peces cebra ha dado lugar a varios compuestos para tratar epilepsias genéticas, entre los que se encuentra el ensayo clínico Argus. A continuación, el Dr. Vicente Villanueva compartió con los asistentes datos de la molécula, resultados del estudio de fase 2 y el nuevo ensayo clínico de fase 3 llamado Skyline.

Posteriormente, el Dr. Rubén Hernández, desde CIMA Universidad de Navarra, nos dio una lección magistral sobre las diferentes estrategias de terapias avanzadas para el síndrome de Dravet. En su presentación incluyó hasta 10 estrategias diferentes de terapias avanzadas para el síndrome de Dravet, las cuales podrían ser un gran avance para esta enfermedad. Además, Jordane Dimidschstein, COO de Regel Therapeutics, nos presentó su tecnología de terapia génica para el síndrome de Dravet y los prometedores resultados preclínico obtenidos, siendo capaces de normalizar los niveles de expresión de SCN1A.

Cerramos el día de nuevo, con nuestra Directora Científica, la Dra. Vittoria Spinosa, en la que nos recalcó el poder y el papel importante de los grupos de pacientes para el desarrollo de nuevos fármacos.

Tras una discusión abierta sobre el futuro del síndrome de Dravet, finalizamos esta estimulante jornada, que nos permitió actualizarnos en el diagnóstico, manejo, terapias disponibles y tratamientos en desarrollo, así como la discusión y el intercambio de ideas para continuar con nuestro objetivo común, la mejora de la calidad de vida de las personas y familias con síndrome de Dravet.

TERMS & CONDITIONS

The Dravet Syndrome Conference 2023 is organised by Dravet Syndrome Foundation Spain, whose registered office is located at Calle Doctor Fleming, 30, 28036 Madrid, Spain.

The conference shall take place at the Official College of Nursing of Madrid, at Avenida de Menéndez Pelayo, 93, 28007 Madrid, Spain, on Thursday 23rd of March 2023. Attendance to this conference is free of charge, but mandatory for organisational reasons.

Participants must register online at www.dravetconference.com. Upon registration, each participant will receive an electronic confirmation by email. On-site registration will not be be possible. If a participant has not received the official email registration confirmation within 3 days, they must contact the conference secretariat at [email protected].

The conference registration includes: Attendance to all sessions, oral and visual presentations, all coffee and lunch breaks, and conference documentation with all information about the event. The registration does not include travel and accommodation costs or any other personal expenses.

Admittance to the conference without registration cannot be granted.

Conference Attendance
Upon arrival at the Official College of Nursing of Madrid the day of the event, all participants have to check-in at the onsite Conference Registration Desk. The desk will open on 23rd of March at 8:30 a.m. Upon registration onsite, the conference team will provide participants with information about the event and a badge, which should be worn visibly at all times. The personalised badge authorises participants to attend all conference sessions, and lunch and coffee breaks.

Cancellation Policy
Dravet Syndrome Foundation Spain reserves the right to cancel any registration forthwith and without liability on its part in the event of any damage to, or destruction of the venue by fire, shortage of labour, strikes, industrial unrest, public health measures, or any other cause beyond the control of the Dravet Syndrome Foundation Spain, which shall prevent it from performing its obligations. In these circumstances, every effort will be made to find an alternative format/venue or propose a new date for the conference. Should the conference be cancelled for any reason, airfares, hotels, or other costs incurred by the participants are not reimbursed by the Dravet Syndrome Foundation Spain.

Any participant is allowed to cancel his/her registration but must notify this to the conference organisers by e-mail at [email protected] within a minimum of 10 days prior the event. Failure to attend the event without prior written notification by email may incur a no-show fee, which shall cover individual organizational, material and catering costs.

Name Change
All registrations are only valid for the respective person mentioned in the form. A registered participant unable to attend the conference may nominate (free of charge) a substitute participant by notifying the conference organisers by email at [email protected] before 16th of March 2023. The procedure allows giving the last name, first name, and email address of the new participant.

Personal Data
Upon registration, the participant authorises Dravet Syndrome Foundation Spain to use their personal data stated above to inform the participant of any further conferences, events, or information concerning the conference and Dravet Syndrome Foundation Spain’s activities. The conference booklet online will only contain authors, titles, and affiliations of submitted abstracts, but no full abstract texts. The conference printed booklet will contain full abstract texts from all participants. Conference certificates will be sent to the participants as a PDF document upon request at [email protected]. Only the name & affiliation will be stated in the certificate, not the email addresses of the participants.

Information is collected by an automated computer system that manages registrations. Security and confidentiality are guaranteed by state-of-the-art technical tools. However, Dravet Syndrome Foundation Spain declines any responsibility concerning data security transmitted through the internet.

Photography
The conference is held in public; therefore, we do not ban participants, exhibitors, partners and other companies from taking photographs of conference activities and participants, and share it via social media. By participating in the conference, you approve that general pictures will be taken and used for our Social Media output, our websites and other communication purposes. You hereby waive any right to oppose, and expressly authorise the organiser to make them available on the Internet or as it may seem fit. Author’s presentation slides shall not be photographed by any conference participant. 

Disclaimer
The conference organiser cannot be held responsible for any loss, injury or damage to any property, whatever the cause may be.
