European Dravet Syndrome Conference 2025: Resumen del día 2 y reflexiones finales

Hoy, 4 de abril de 2025, hemos celebrado el segundo y último día de la European Dravet Syndrome Conference en el Colegio Oficial de Enfermería de Madrid, cerrando dos jornadas llenas de ciencia, debate y esperanza para la comunidad del síndrome de Dravet.

Mecanismos de acción: Explorando tratamientos innovadores

La jornada comenzó con una sesión dinámica sobre el mecanismo de acción de diversos tratamientos para las encefalopatías epilépticas y del desarrollo, familia a la que pertenece el síndrome de Dravet. Liderada por el Dr. Massimo Mantegazza, esta sesión destacó por su interactividad: las preguntas del ponente a los asistentes generaron un debate enriquecedor sobre estrategias de terapias avanzadas y de precisión, abriendo nuevas perspectivas para el futuro.

Avances en terapias génicas: Un horizonte prometedor

A continuación, tuvimos una sesión centrada en la investigación, moderada por la Dra. Heidrun Potschka. En esta sesión pudimos escuchar charlas de investigación pre-clínica sobre nuevas posibles estrategias de terapias génicas para el síndrome de Dravet. La doctoranda Elena Benvenuto presentó su estudio sobre el rescate de la haploinsuficiencia de SCN1A mediante la mejora de la traducción, es decir, el proceso que va desde el gen hasta la proteína Nav1.1, mostrando un nuevo enfoque de edición de bases. Luego, la Dra. Samantha Hanson expuso su investigación sobre la durabilidad de la eficacia de una terapia génica específica para interneuronas en un modelo de ratón con síndrome de Dravet, mostrando resultando esperanzadores. Por su parte, el Dr. Juan Antinao-Díaz compartió avances en selección génica con un vector viral que incluye un transcrito antisentido natural de SCN1A, planteando una posible nueva vía de tratamiento.

Nuevos enfoques en investigación: Biomarcadores y modelos celulares

Tras un breve receso para el café, tuvimos la tercera sesión centrada en la investigación, moderada por el Dr. Franck Kalume, miembro de nuestro comité científico. El doctorando Georgii Krivoshein presentó su estudio en modelos animales sobre un posible marcador de electroencefalograma (EEG) llamado delta-gamma PAC (θ-γ PAC) para evaluar la eficacia de los fármacos anticrisis. Luego, el doctorando Oskari Kulta expuso su investigación sobre las anomalías en la actividad funcional de las redes neuronales en un modelo humano de células inducidas pluripotentes de síndrome de Dravet. Este es un modelo celular que deriva de células de pacientes y que nos permite mimetizar la mutación de un paciente in-vitro. Finalmente, la Dra. Tiffany Migevent, desarrolló sus avances en los mecanismos y la terapia de la muerte súbita e inesperada en epilepsia (SUDEP) en un modelo murino de encefalopatía epiléptica y del desarrollo, que es la gran familia de enfermedades en la que se encuadra el síndrome de Dravet.

Modelos preclínicos: Herramientas para el progreso

Continuamos con una sesión interactiva dirigida por el Dr. Thomas Wuttke sobre los modelos preclínicos in-vitro e in-vivo para el estudio de encefalopatías epilépticas y del desarrollo, incluyendo modelos de células pluripotentes inducidas y organoides, así como modelos de roedores y peces cebra. Esta sesión generó numerosas preguntas entre los asistentes sobre las limitaciones y las ventajas de dichos modelos. Todos los asistentes que intervinieron concordaron en que estos modelos son necesarios y que nos han permitido y nos permiten hacer grandes avances en la mejora del conocimiento del síndrome de Dravet y otras enfermedades similares.

Cribado neonatal: Oportunidades y dilemas éticos

La sesión siguiente fue un panel de expertos moderado por la Dra. Rima Nabbout, centrado en el cribado neonatal (prueba del talón) y la intervención presintomática. Contamos con la presencia de distinguidos ponentes como Antonio Gil-Nagel, neurólogo especialista en epilepsia; Naomi Fukuda, representante de la asociación japonesa de pacientes con síndrome de Dravet; el Dr. Rubén Hernández, investigador experto en terapia génica; y la Dra. Astrid Pañeda Rodríguez, directora senior Forge Biologics. El cribado neonatal podría ser una forma de detectar el síndrome de Dravet en etapas neonatales, poco después del nacimiento y antes de la aparición de los síntomas. Se ha hablado de la posibilidad de un tratamiento temprano, cómo éste podría modificar los síntomas del paciente y de cuál sería el momento más adecuado para hacerlo. Sin embargo, tras una discusión muy intensa con la audiencia, se han expresado preocupaciones sobre los aspectos éticos de comunicar el resultado de un cribado neonatal previo a la aparición de síntomas, ya que hasta el momento no se puede asegurar que una mutación en el gen SCN1A cause necesariamente el síndrome de Dravet y que, además, de momento no tenemos tratamientos adecuados que curen la enfermedad, aunque para ello trabajamos. Naomi Fukuda nos ofreció la perspectiva de los pacientes de Japón, donde se vio que alrededor de la mitad de ellos estaban satisfechos con el tiempo del diagnóstico, aunque este a veces superara los dos años. Un 30% de las familias no querrían ser incluidas en un cribado neonatal porque “no cambiaría la realidad”, “crearía ansiedad sin dar soluciones”. Sin embargo, cuando se preguntó a los asistentes de nuestra conferencia a mano alzada si se debería incluir el síndrome de Dravet en los cribados neonatales, alrededor del 60% estaba a favor de ello.

Vectores virales y análisis transcriptómico: Nuevas fronteras

Después de la comida, tuvimos una nueva sesión dirigida por el Dr. Eric Kremer sobre los vectores virales caninos y su uso para un posible tratamiento para el síndrome de Dravet. Finalmente se realizó la cuarta y última sesión de investigación, moderada por la Dra. Rajvinder Karda. Nuestra directora científica, la Dra. Simona Giorgi, presentó un estudio sobre el análisis transcriptómico de sangre periférica en personas con síndrome de Dravet, que la Fundación Síndrome de Dravet realizó gracias a la colaboración de los que participasteis en este estudio en nuestra Reunión Anual de Familias de 2024. A continuación, la Dra. Rima Nabbout cerró la sesión con una exposición sobre las diferentes perspectivas de una investigación etnográfica sobre el síndrome de Dravet, una investigación donde se han analizado los diferentes perfiles de cuidador.

Reconocimiento y cierre

Para concluir, nuestro presidente, José Ángel Aibar, y la Dra. Ingrid Scheffer entregaron los premios al mejor póster a Martina Mainardi, de la Universidad Vita-Salute San Raffaele, y a la mejor presentación oral a Ellie Chilcott, del University College de Londres.

Un legado de unión y esperanza

Estos dos días de la European Dravet Syndrome Conference 2025 han sido un viaje inspirador a través de la ciencia, la empatía y la colaboración. Desde las bases clínicas y diagnósticas del primer día hasta las innovaciones terapéuticas y los debates éticos del segundo, hemos construido un espacio donde clínicos, investigadores, profesionales de la industria farmacéutica y representantes de pacientes han unido fuerzas para transformar el futuro del síndrome de Dravet. Las conexiones forjadas y las ideas compartidas nos impulsan con determinación hacia un objetivo común: mejorar la vida de quienes enfrentan esta enfermedad y, algún día, hallar una cura. Gracias a todos los que han hecho posible este encuentro; el camino sigue adelante, y lo recorremos juntos.

TERMS & CONDITIONS

The Dravet Syndrome Conference 2023 is organised by Dravet Syndrome Foundation Spain, whose registered office is located at Calle Doctor Fleming, 30, 28036 Madrid, Spain.

The conference shall take place at the Official College of Nursing of Madrid, at Avenida de Menéndez Pelayo, 93, 28007 Madrid, Spain, on Thursday 23rd of March 2023. Attendance to this conference is free of charge, but mandatory for organisational reasons.

Participants must register online at www.dravetconference.com. Upon registration, each participant will receive an electronic confirmation by email. On-site registration will not be be possible. If a participant has not received the official email registration confirmation within 3 days, they must contact the conference secretariat at [email protected].

The conference registration includes: Attendance to all sessions, oral and visual presentations, all coffee and lunch breaks, and conference documentation with all information about the event. The registration does not include travel and accommodation costs or any other personal expenses.

Admittance to the conference without registration cannot be granted.

Conference Attendance
Upon arrival at the Official College of Nursing of Madrid the day of the event, all participants have to check-in at the onsite Conference Registration Desk. The desk will open on 23rd of March at 8:30 a.m. Upon registration onsite, the conference team will provide participants with information about the event and a badge, which should be worn visibly at all times. The personalised badge authorises participants to attend all conference sessions, and lunch and coffee breaks.

Cancellation Policy
Dravet Syndrome Foundation Spain reserves the right to cancel any registration forthwith and without liability on its part in the event of any damage to, or destruction of the venue by fire, shortage of labour, strikes, industrial unrest, public health measures, or any other cause beyond the control of the Dravet Syndrome Foundation Spain, which shall prevent it from performing its obligations. In these circumstances, every effort will be made to find an alternative format/venue or propose a new date for the conference. Should the conference be cancelled for any reason, airfares, hotels, or other costs incurred by the participants are not reimbursed by the Dravet Syndrome Foundation Spain.

Any participant is allowed to cancel his/her registration but must notify this to the conference organisers by e-mail at [email protected] within a minimum of 10 days prior the event. Failure to attend the event without prior written notification by email may incur a no-show fee, which shall cover individual organizational, material and catering costs.

Name Change
All registrations are only valid for the respective person mentioned in the form. A registered participant unable to attend the conference may nominate (free of charge) a substitute participant by notifying the conference organisers by email at [email protected] before 16th of March 2023. The procedure allows giving the last name, first name, and email address of the new participant.

Personal Data
Upon registration, the participant authorises Dravet Syndrome Foundation Spain to use their personal data stated above to inform the participant of any further conferences, events, or information concerning the conference and Dravet Syndrome Foundation Spain’s activities. The conference booklet online will only contain authors, titles, and affiliations of submitted abstracts, but no full abstract texts. The conference printed booklet will contain full abstract texts from all participants. Conference certificates will be sent to the participants as a PDF document upon request at [email protected]. Only the name & affiliation will be stated in the certificate, not the email addresses of the participants.

Information is collected by an automated computer system that manages registrations. Security and confidentiality are guaranteed by state-of-the-art technical tools. However, Dravet Syndrome Foundation Spain declines any responsibility concerning data security transmitted through the internet.

Photography
The conference is held in public; therefore, we do not ban participants, exhibitors, partners and other companies from taking photographs of conference activities and participants, and share it via social media. By participating in the conference, you approve that general pictures will be taken and used for our Social Media output, our websites and other communication purposes. You hereby waive any right to oppose, and expressly authorise the organiser to make them available on the Internet or as it may seem fit. Author’s presentation slides shall not be photographed by any conference participant. 

Disclaimer
The conference organiser cannot be held responsible for any loss, injury or damage to any property, whatever the cause may be. If for any reason the organiser decides to make material changes to this conference, the organiser is not responsible for airfares, hotels or other costs incurred by the participants. The participant takes part in the conference, all tours, and trips at his own risk.
